in the development and validation of therapeutic agents for cancer and neurological disorders, and a range of other diseases.1,2,3 Reported studies have used numerous agents including 18FDG, 18FLT, 18FMISO and 18FAZA as early biomarkers of drug efficacy.2,4 Preclinical PET imaging has been proven in studies of therapeutic targeting and biodistribution, drug target interactions, pharmacodynamics and toxicology.5 These methods have helped facilitate the development of therapeutic agents that are now clinically available.6 Probably two of the most common uses of preclinical PET imaging in drug discovery are studies of compound targeting and biodistribution and therapeutic efficacy. Here, we will provide a brief introduction to preclinical PET imaging for such studies. Targeting and Biodistribution

Objective(s): Gallium-68 DOTA-DPhe1-Tyr3-Octreotide (68Ga-DOTATOC) has been applied by several European centers for the treatment of a variety of human malignancies. Nevertheless, definitive dosimetric data are yet unavailable. According to the Society of Nuclear Medicine and Molecular Imaging, researchers are investigating the safety and efficacy of this radiotracer to meet Food and Drug Admin...
متن کاملDevelopment of positron emission tomography β-amyloid plaque imaging agents.
For 100 years, β-amyloid (Aβ) plaques and neurofibrillary tangles (NFTs) have been recognized as the neuropathological hallmarks of Alzheimer's disease (AD), and their presence or absence could only be assessed postmortem using stains and dyes that identified these microscopic structures. Approximately 10 years ago, the first successful Aβ plaque-specific positron emission tomography (PET) imag...
متن کاملPET and MRI-guided focused ultrasound surgery for hypoxic-tissue ablation combined with radiotherapy in solid tumors
Background: The rationale was to develop an ablation approach to destroy regions of tumor resistant to radiation and thus reduce the time required for whole tumor ablation, while improving overall tumor control after radiotherapy. Materials and Methods: The system is composed of a micro positron emission tomography (mPET), 7T magnetic resonance imaging (MRI), and a customized MRI-compatible foc...
متن کاملEarly detection of Alzheimer's disease with PET imaging.
Preclinical diagnosis of Alzheimer's disease (AD) is one of the major challenges for the prevention of AD. AD biomarkers are needed not only to reveal preclinical pathologic changes, but also to monitor progression and therapeutics. PET neuroimaging can reliably assess aspects of the molecular biology and neuropathology of AD. The aim of this article is to review the use of FDG-PET and amyloid ...
متن کاملThe effect of fasting on Positron Emission Tomography (PET) imaging
As a nuclear approach, Positron Emission Tomography (PET) is a functional imaging technique which is based on the detection of gamma ray pairs emitted by a positron-emitting radionuclide. There are certain limitations to this technique such as normal tissue uptake. Therefore, it has been recommended that patients prepare before scanning. Fasting for a short while before PET imaging is an exampl...
